I am switching to Plusnet on Thursday. The new router (Plusnet Hub One) has arrived. My PC is currently connected by ethernet cable and all other devices by wifi. The instructions to Hub One say I have to unplug the ethernet cable when switching to wifi. Does this mean that I cannot use wifi and ethernet connections at the same time?

Welcome to the forum.
No it's telling you not to use both on the same device. You can run a mixture of wired and wireless devices at the same time as with any router.
