Simple question !

I opted to change into the Plusnet home phone without really looking at it to be honest.
I notice that my balance on the account is £50.00, does this mean that i can use £50.00 worth of free minutes off peak, and after this has gone i start getting charged?

no that is a fall back for chargeable call, i.e out side the free period, over 60 mins, mobile calls, 0870, 0845 calls, these are normally added to your bill at the end of the month thus leaving you with the£50 limit

Ok, so if my balance is -0.47p.
That means i have made 47p worth of peak calls?

something like that, if you go to the members centre, phone calls, view, Itemised bill it is here it will show you  you can also click details to get more info

It means you've made 47p worth of *chargeable* calls - i.e. daytime calls (assuming you're on Evenings and Weekends), charegeable part of calls over over 1 hour, calls to mobiles, 0845, 0870, etc.
