I have a Mac mini that runs in headless mode and goes to sleep when not in use.
On previous router (sky supplied sr101) i could wake up the mac mini simply by trying to access it via the network.
Now when using the Technicolor TG582n, the mac mini doesn't appear to wake up.
So, is WOL not supported in this router, or does it need to be manually configured to do so?

I fixed that by disabling the firewall in the router - and I have no idea why that worked but it may be worth a try

i have no issues seeing local LAN once a device awakes from sleep.
also, you don't want to be disabling the firewall, as that allows all incoming connections to come thru unfiltered.

any previous router i've used did not require a specific rule to be set up and then configured for an individual device.... it "just worked" with no manual config.
i need it to work for any device that is not the network.

If you need to wake the pc from the lan then you don't need to do anything with the Technicolor router, it should just work.
If you wish to wake a number of devices at the same time from the internet then replace the device MAC address, in the above link, with a broadcast MAC address.
Don't remember the details but should be easy to find from google.

mac mini is not being woken up by WOL requests.
the mac mini is still visible as a device on the network when sleeping (as it should), but the WOL request is obviously not being sent to the mac mini.

Put another machine on the LAN and run a packet sniffer like Wireshark to check that the WOL packet is being correctly sent out.
You could also try temporarily putting your Sky router back in play (it doesn't need connecting to the Internet) and repeating the above tests.

i don't have another machine to put on the LAN, just the mac mini, and the macbook pro that i'm using to try and wake it up (via screen sharing).

The test is only to verify that the magic packet is at least reaching the machine.
